Description

Sofovir (Sofosbuvir 400mg) is used with other medications in the treatment of chronic (long-lasting) hepatitis C virus (HCV) infection in adults. It is an antiviral medicine that lowers the amount of virus in the body and works to eliminate the virus over a period of time.

Pharmacodynamics 

Sofosbuvir tends to inhibit the Hepatitis C NS5B protein. It shows a high barrier to the development of resistance. Sofosbuvir is a prodrug of the Protide type, wherein the active phosphorylated nucleotide is granted with cell permeability and oral bioavailability. By hydrolysis of the carboxylate ester by either of the enzymes cathepsin A or carboxylesterase 1, Sofosbuvir is activated in the liver to the triphosphate GS-461203, which is followed by cleaving of the phosphoramidate by the enzyme histidine triad nucleotide-binding protein 1 (HINT1), with repeated phosphorylation. Dephosphorylation tends to create the inactive metabolite GS-331077. The half life of Sofosbuvir is 0.4 hours, and the half life of GS-331077 is 27 hours. Following a single 400 mg oral dose of Sofosbuvir, 80% is normally excreted in urine, 2.5% in expired air recovery and 14% in feces.
