Hepcinat (Sofosbuvir) 400mg tablet is an antiviral medication, used along with other medications, in the treatment of chronic Hepatitis C Virus (HCV) infection in adults. This medicine works to lower the growth and multiplication of virus in the body and helps to eliminate the virus from the body.
Sofosbuvir tends to inhibit the Hepatitis C NS5B protein. It shows a high barrier to the development of resistance. Sofosbuvir is a prodrug of the Protide type, wherein the active phosphorylated nucleotide is granted with cell permeability and oral bioavailability. By hydrolysis of the carboxylate ester by either of the enzymes cathepsin A or carboxylesterase 1, Sofosbuvir is activated in the liver to the triphosphate GS-461203, which is followed by cleaving of the phosphoramidate by the enzyme histidine triad nucleotide-binding protein 1 (HINT1), with repeated phosphorylation. Dephosphorylation tends to create the inactive metabolite GS-331077. The half life of Sofosbuvir is 0.4 hours, and the half life of GS-331077 is 27 hours. Following a single 400 mg oral dose of Sofosbuvir, 80% is normally excreted in urine, 2.5% in expired air recovery and 14% in feces.
